You need to choose the "Settings..." command from the Options menu in Project64. On the Plugin tab of the Settings dialoge that comes up, are all three plugin fields filled in? If not, fill them in with these plugins:
Graphics/Video: Jabo"s Direct3D 8 Sound/Audio: Jabo"s DirectSound 1.6 Input/Controller: Jabo"s DirectInput 1.6

If the field is blank and you have nothing to fill it in with, that means you must have deleted some of your plugins, or maybe even the entire Plugin folder of Project64.
Try reinstalling Project64, and then it should work.

Just choose Repair, and click finish. Then run Project64.exe, Options/Settings, make sure all three plugin fields are filled in, click OK, and then the File/Open to select a ROM should work.
